I set my Raspberry Pi Zero W to be a DNLA WiFi Server but connecting a USB flash to it and use VLC but it shows the USB flash to be empty and there's video files in it

I take it you mean that you've enabled UPnP sharing? That will only apply to videos/music that are scraped into the Kodi Libraries.
Multimedia files located elsewhere will not be shared.

Enable the Samba server in LE settings.
